Absorp-tion of a photon with energy above the semi-conductor band gap energy results in thecreation of an electron-hole pair (or exciton).The absorption has an increased probabilityat higher energies (i.e., shorter wavelengths)and results in a broadband absorptionspectrum, in marked contrast to standardfluorophores (Fig. 1B). For nanocrystalssmaller than the so-called Bohr excitonradius (a few nanometers), energy levels arequantized, with values directly related to theqdot size (an effect called quantum confine-ment, hence the name ‘‘quantum dots’’) (4).The radiative recombination of an exciton[characterized by a long lifetime, 910 ns (5)]leads to the emission of a photon in a narrow,symmetric energy band (Fig. 1), another dif-ference from the red-tailed emission spectraand short lifetimes of most fluorophores. Thelong fluorescence lifetime of qdots enablesthe use of time-gated detection (6)toseparate their signal from that of shorterlived species (such as background autofluo-rescence encountered in cells).Surface defects in the crystal structure actas temporary ‘‘traps’’ for the electron or hole,preventing their radiative recombination. Thealternation of trapping and untrapping eventsresults in intermittent fluorescence (blinking)visible at the single-molecule level (7) andreduces the overall quantum yield, which isthe ratio of emitted to absorbed photons. Oneway to overcome these problems, and toprotect surface atoms from oxidation andother chemical reactions, is to grow a shell ofa few atomic layers of a material with alarger band gap on top of the nanocrystalcore. This shell can be designed carefully toobtain quantum yields close to 90% (8); thisstep also enhances qdots’ photostability byseveral orders of magnitude relative toconventional dyes (9).Single qdots can be observed and trackedover an extended period of time (up to a fewhours) with confocal microscopy (10), totalinternal reflection microscopy (11, 12), orbasic wide-field epifluorescence microscopy(12–14).
